What is the Intex system?The Intex-Allergoset is a unique test system
for the determination of total serum IgE aswell as circulating IgE specific for food and inhalant allergens. Allergic diseases, which are influenced by both genetic and environmental factors, are
characterized by the formatin of IgE antibodies after sensitisation and repeated exposure to certain allergens. Thus, the detection of allergen specific IgE antibodies facilitates the identification of allergy causing
substances.
This system is an enzyme immunoassay-based dipstick test
system that is carried out in special blisters. After the immunological reaction of serum IgE with its solid phase ligand (anti IgE for total IgE, and different allergen for specific IgE) the
bound IgE is detected by immunoperoxidase method. The food allergen-specifiic IgE in the sample (serum or
heparinised plasma) is allow to react with different food allergens bound to the solid phase (dipstick). After washing to remove unbound material, the dipstick is
incubated with peroxidase-labelled monoclonal anti-IgE antibody (Conjugate IgE sp.) Excess reagent is then removed by washing, and the dipstick is placed in an enzyme-substrate solution. The peroxidase reaction
is visualised by a chromogen.
The developed colour intensity is directly proportional to the
concentration of food allergen-specific IgE in the sample. The results may be evaluated photometrically ( in a densitometer) and expressed in INTEX-classes (titer units) from 0 to 4.
Each dipstick comprises two internal control dots, an IgE-positive value to indicate that the test has performed properly and a negative control which is the base point for the calculation of titers. Outline of the exact Procedure involved
