1. Preparatory Steps
- Bring all reagents and sample to room temperature (RT)
- Tear open blister card and remove strip
- Cut blister as shown
- Mark strip and fill in the patient card

2. First immunological reaction
- Fill blister no. 1 to the marking (1) with serum or heparinized plasma
- Place strip into blister
- remove air bubbles by vertical movements of the dipstick
- Incubate overnight (16-24 h) at RT

3. First wash
- Incubate strip in distilled water at RT for 10 minutes
- Remove water by gently blotting with a paper towel
|
4. Second immunological reaction
- Fill blister no.2 to the marking (1) with Conjugate IgE sp.
- Place strip into blister
- Remove air bubbles by vertical movements of the strip
- Incubate 1hr at RT

5. Second Wash
- Incubate dipstick in distilled water 2 X 10 minutes at RT
- Remove water by gently blotting with a paper towel

6. Enzyme Reaction
- Fill blister no. 3 to the first marking (1/6) with Chromagen.
- Add a Substrate Buffer to the second marking
- Place strip into blister and mix solutions by vertical movements of the dipstick

7. Third Wash
- Incubate strip in distilled water for 5 minutes at RT
- Remove water by gently blotting with a paper towel
- Dry strip in the dark for 30 minutes at RT

8. Evaluation of Resluts by Color Card or Densitometer
- Determine INTEX Class by comparison with the color card, or use the Densitometer
- Introduce results into a patient card.
